Regulation of calcium ion level by vasoactive intestinal peptide in human umbilical vein endothelial cells in vitro

Abstract
AIM:To observe the regulation on intracytosolic free calcium ion and its membrane channel of human umbilical vein endothelial cells(HUVEC) by vasoactive intestinal peptide(VIP). METHODS: Confocal laser scanning microscopy was used to observe the concentration of intracytosolic free calcium ion and patch-clamp technique to detect opening probability of calcium ion channel in the membrane of HUVEC cultured in vitro. RESULTS:VIP could increase the opening probability(1.1% to 6.2), prolong the opening period(0.304 ms to 0.706 ms),shorten the closing period(14.019 ms to 9.481 ms) of calcium ion channel in HUVEC from in vitro.The fluorescence intensity of HUVEC intracytosolic free Ca2+ cultured in vitro increased obviously by VIP,it began to change in the 30 s after VIP was added, which showed that Ca2+ increased more significantly than that in the control group. CONCLUSION: VIP may function as the activator of the increase in the intracytosolic free calcium ion in the cultured HUVEC in vitro,and the inflowing of calcium ion from the outside of cell bodies.关键词
