Abstract
BACKGROUND: Lipid peroxidation results in disturbance of learning and memory, but radix echinopsis inhibits lipid peroxidation and acts on improving immune function of cell and resisting peroxidation of free radical.OBJECTIVE: To probe into the effects of alcohol extract of radix echinopsis on learning and memory of senile mice induced by D-galactose through the changes in lipid peroxide (LPO) and lipofuscin contents.DESIGN: Randomized controlled experiment was designed.SETTING: Teaching-Research Room of Pharmacology in School of Pharmacy of Shenyang Pharmaceutical University, Teaching-Research Room of Chinese Medicine of the First Clinical College of China Medical University, Teaching-Research Room of Natural Medicine and Chemistry in School of Traditional Chinese Medicine of Shenyang Pharmaceutical University.MATERIALS: The experiment was performed in Teaching-Research Room of Pharmacology in School of Pharmacy of Shenyang Pharmaceutical University from February 1994 to October 1994. Totally 185 Kunming mice of clean grade were employed and randomized into 7 groups, named alcohol extract of radix echinopsis 3.125 g/(kgod) group [3.125 g/(kgod)group]](30 mice), alcohol extract of radix echinopsis 1.56 g/(kgod) group [1.56 g/(kgod) group] (30 mice), alcohol extract of radix echinopsis 0.78 g/(kgod) group [0.78 g/(kgod) group] (30 mice), alcohol extract of radix echinopsis 0.39 g/(kgod) group [0.39 g/(kgod) group] (15 mice), piracetam group (15 mice), blank control (30 mice) and model group (35 mice).METHODS: Alcohol extract of radix echinopsis was infused in each of the groups of alcohol extract of radix echinopsis at 3.125, 1.56, 0.78 and 0.39 g/kgod successively. In piracetam group, piracetam 0.2 g/kgod was applied. Distilled water of equal volume was infused in blank control and model group respectively. Except black control, in rest groups, 50 g/L Dgalactose was injected subcutaneously everyday, 0.5 mL/mouse, continuously for 40 days, by which, senile mouse model was prepared. Forty days later, learning and memory were determined in each group with step down test and step through test.groupRESULTS: Totally 185 mice were employed in the experiment, totally,group: compared with model group, the error in 3.125,1.56,0.78 g/(kgod)groups was reduced remarkably [( 1.68±1.25 ), (0.92±0.73), (0.78±Changes in errors and latent phase in step through test in each group:compared with model group, the error in 3.125,1.56,0.78 g/(kgod)groups was reduced remarkably(P < 0.05 or P < 0.01) successively and the laof LPO and lipfuscin contents in brain tissue of each group: compared with model group, those in 3.125,1.56,0.78 g/(kgod)groups were reduced remarkably successively(P < 0.05 or P < 0.01).CONCLUSION: Alcohol extract of radix echinopsis acts on resisting aging, benefiting intelligence, being against peroxidation of free radical and reduces the production of LPO and lipofuscin in brain tissue, which is probably attributed to its action on improving memory disturbance.关键词
