Abstract
[Objective] To investigate Bcl-w expression and its relationship with clinical pathologic factors in colorectal carcinoma. The effect of Bcl-w expression on the apoptosis and the relationship with Bcl-2 and P53 expression were also detected in these tumors.[Methods] The expression of Bel-w, Bel-2 and P53 was studied in 52 colorectal carcinoma tissue samples by using immunohistochemistry. Bel-w expression was further assessed with Western blot. The function of Bcl-w expression on the apoptosis of colorectal carcinoma was detected by TUNEL. [Results] Positive immunoreactivity was identified of Bcl-w in 39 of 52(75%) colorectal cancer specimens. The expression level of Bcl-w was increased with tumor's invasion, lymph node and liver metastasis.Patients with tumor expressed Bcl-w were all died within 3 years after surgery. In contrast to Bcl-2, colorectal adenoma had not expressed Bcl-w, the statistics analysis revealed a significant negative correlation between the Bcl-w and apoptosis and a clear inverse relationship between P53 and Bcl-w expression.[Conclutions] The expression of Bcl-w is significantly related to the progression of the colorectal cancer. It could be regarded as a prognostic factor for this cancer. In contrast to Bcl-2, Bcl-w play an important role in the advanced tumor stage. Furthermore, Bcl-w is a potent suppressor for apoptosis in colorectal carcinoma. An effect of P53 on down-regulating Bel-w expression is also demonstrated.关键词
