Abstract
[Objective] HCCR1 and HCCR2 are the oncogenes overexpressed in hepatocellular carcinomas(HCC). In this experiment, it was evaluated that the cloning, bacterial expression, purification and characterization of soluble recombinant proteins of HCCR1 and HCCR2 for novel early diagnosis of HCC. Protein solublity was key to producing recombinant proteins for founction determination. [Method] Five methods, cDNA clone and expression,protein purification, solubility assay and western blod assay were used to evaluate production of HCCR1 and HCCR2 proteins, enzyme-linked immunosorbent assay (ELISA) was used to detect HCCR1 and HCCR2 antigens in serum.[Result] The results show that the proteins of HCCR1 and HCCR2 were expressed in Escherichia coli at high levels. [Conclusion] The recombinant proteins are soluble and may used to generate the antibody for early diagnosis of HCC. Finally, HCCR1 and HCCR2 may be useful in monitoring HCC, and as potential disease specific therapeutic targets.关键词
