牛心包纤维支架细胞重建组织工程体外实验研究
Experimental research on tissue engineering for cellular reconstitution on pericardial fibrous scaffold in vitro

Abstract
To construct a tissue-engineering material of bioprostheses. Methods: Group Ⅰ: fresh bovine pericardium was acellularized with Courtman's method and fixed with glutaraldehyde (GA); Group Ⅱ: first treated as Group Ⅰ, then was modified with 2, 3-butanediol. Myofibroblast cells(MFC) and endothelial cells(EC) were isolated from porcine carotid artery separately, then cuhured in vitro, and seeded sequentially.(3) For analysis of specimens, Trypan blue staining, Hematoxylin-eosin (HE) staining, Ⅷ-factor andα-actin staining,Scanning electron microscope (SEM) and transmission electron microscope (TEM) were performed. Results: Courtman's method could get rid of most of the pericardial cells, the matrix structure became a biological fibrous scaffold which was lightly loosed with many fissures. No cells grew in group Ⅰ ,the cells of group Ⅱ proliferated after seeding; On the 17th day,some MFC grew into the matrix in Group Ⅱ; On the 27th day, 2-3 layers of cells grew on the pericardial fibrous scaffold in group Ⅱ, cells on the surface were EC, in the matrix were MFC, and EC was an intact confluent monolayer, some of the EC were spindle-shaped. Conclusions: Biological fibrous scaffold which is from acellular, fixed and modified bovine pericardium has excellent biocompatibility. Separate isolating, separate culture and separate seeding of MFC and EC are practical tissues engineering technique for the re-cellularization on biological fibrous scaffold, and this scaffold can be completely endothelialized and partly interstitial cellularized in vitro.关键词
