Abstract
Aim To observe the effect of glucocorticoids on marrow stromal cell proliferation and differentiation and study the pathological mechanism of osteoporosis.Methods Bone marrow matrix cells were divided into experiment and control groups according to random number table.Proliferation of marrow stromal cell treated with dexamethasone(1× 10- 7 mol/L) and contents of osteocalcin in the media were measured.Results The number of the marrow stromal cell[(3.73± 0.43)× 105]and the contents of osteocalcin[(5.2± 1.4)ng/mg]in the media of the experimental group obviously decreased compared to the control[(5.76± 0.69)× 105,(12.5± 1.5)ng/mg](t=12.61,7.45,P< 0.001).The number of adipocytes increased with longer duration of exposure to dexamethasone.Conclusion Glucocorticoids inhibited marrow stromal cell proliferation and osteoblastic differentiation, and promoted its differentiation into adipocytes,which is a major contributing factor of bone loss and an increase in marrow fat volume in the osteoporosis.关键词
