人肝癌特异性EB病毒载体-pEBAF介导hGM- CSF基因的转移及表达

Abstract
Objective: To investigate a new way of tissue-specific expression of human granulocyte-macrophage colony stimulating factor (hGM-CSF) gene in HCC. Methods: Construct pEBAF/hGF- CSF, transferred into HCC cell line HepG2 (secretingAFP) and SMMC7721 (not secreting AFP) by lipofectin,two positive cell lines HepG2/hGM- CSF and SMMC7721/hGM-CSF.Results:By MTT assay the cytokine biological activity revealed the HepG2/hGM-CSF expressed hGM-CSF for 46.5ng/ml/l06/24hr,but SMMC7721 has no expression of hGM - CSF were established. Conclusions: hGM- CSF gene mediated by pEBAF has theablility of tissue- specific expression in HCC cells.关键词
