Abstract
AIM:To evaluate the ability of bone morphogenetic protein 4 (BMP4) in inducing the in vitro differentiation of adult rat bone marrow-derived mesenchymal stem cells (MSCs) into neurons. METHODS:Using immunohistochemichal method,the expressions of neurofilament protein(NF) and glial fibriliary acidic protein (GFAP) were detected in BMP4-induced MSCs of the fifth passage. RESULTS:BMP4-induced MSCs were transformed into neuron-like cells,which were positive for NF expression as shown by immunocytochemical determination.The percentage of NF-positive neuron-like cells at 5 h,12 h,1 d, 3 d, 5 d and 7 d after induction was (40± 7)% ,(71± 6)% ,(58± 3)% ,(53± 6)% ,(37± 5)% and (13± 2)% respectively.The peak of NF expression in the neuron-like cells occurred at 12 h after induction,which was higher than that 5 h after induction(t=1.380- 2.621,P< 0.05). CONCLUSION:BMP4 can induce rat bone marrow-derived MSCs into neurons in vitro.关键词
