人参对大鼠缺血再灌注后心肌细胞凋亡的抑制作用
Inhibitory effects of Radix ginseng rubra on cardiomyocyte apoptosis induced by ischemia and reperfusion in rats

Abstract
BACKGROUND:Myocardial ischemia and reperfusion injury is the clinical hotspot recently.Whether ginseng and its extract-ginsenoride have the effect on the cardiomyocyte apoptosis induced by ischemia-reperfusion injury is not clear. OBJECTIVE:To observe the influence of Radix ginseng rubra on the cardiomyocyte apoptosis and the expression of bcl-2 gene in the ischemia-reperfusion heart of rat in vivo. DESIGN:Randomized controlled experimental study conducted. SETTING AND MATERIALS:Fifteen Wistar rats(provided by Zoology Department of Tongji Medical University) of both sexes,weighing 200- 500 g were used in the present study.The research was conducted in the Experimental Center of Tongji Hospital,Wuhan,Hubei Province,China. INTERVENTION:The left anterior descending(LAD) coronary artery was ligated to establish the ischemia/reperfusion heart model in Wistar rats.The apoptotic cardiomyocyte was shown by in situ end labeling method and the number of apoptotic cardiomyocyte was counted under optical microscopy. Immunohistochemistry and in situ hybridization histochemistry(ISHH) were used to detect mRNA and protein of bcl-2 gene expression respectively. Image processing system was used to quantify dispose the positive metric substance of both immunohistochemistry and ISHH through the average absorbance(A) value. MAIN OUTCOME MEASURES:Whether Radix ginseng rubra has the effects on the cardiomyocyte apoptosis and the expression of bcl-2 mRNA and Bcl-2 protein in the ischemia-reperfusion heart of rat in vivo. RESULTS:A values of bcl-2(ISHH) were 0.11± 0.02 in Radix ginseng rubra group,0.07± 0.02 in LAD ligation group and 0.06± 0.01 in sham operated group respectively. Bcl-2 gene expression was increased significantly in Radix ginseng rubra group as compared with LAD ligation group and sham-operated group(t=8.72,P< 0.001).While A values of Bcl-2(immunohisochemistry) were 0.15± 0.02(Radix ginseng rubra group),0.14± 0.01(sham operated group),and 0.09± 0.02(LAD ligation group) respectively.A value of Bcl-2 protein was increased significantly in Radix ginseng rubra group as compared with LAD ligation group(t=8.631,P< 0.001),but the difference was not significantly between Radix ginseng rubra group and sham-operated group(t=2.67,P > 0.05). CONCLUSION:Radix ginseng rubra has obviously preventive and therapeutic effects on reperfusion injury and could inhibit cardiomyocyte apoptosis.The mechanism might be that bcl-2 gene expression is increased by ginseng in the cardiomyocyte following ischemia-reperfusion injury.关键词
