Abstract
Objective:To study the effect of breviscapine on intracellular free Ca2+ concentration ([Ca2+]i) induced by KCl in cultured vascular smooth muscle cells (VSMC) of rabbits.Methods:VSMCs were cultured and loaded with Fura-2AM.[Ca2+]i was measured by fluorospectrophotometer (Shimadzu RF_5000,Japan).Results:In could slightly decrease [Ca2+]i in resting status,which had no statistic significance.Depolarization induced by high
KCl 50mmol·L-1 could increase [Ca2+]i to 420±104nmol·L-1(P＜0.05),whereas 10-6、10-5、3×10-5mol·L-1 of breviscapine could further enhance [Ca2+]i elevation induced by KCl 50mmol·L-1(P＜0.05).Neverhteless,breviscapine had no significant effect on [Ca2+]i in the absence of extracellular calcium.Conclusion:Breviscapine can enhance Ca2+ influx into muscle cells through voltage-dependent calcium channels.关键词
