Activation of microglia and recruitment of extraneous macrophage following ischemia-reperfusion in adult rats

摘要

Abstract

AIM: To study the activation and proliferation of microglia/macrophages in cerebral tissue in rats following ischemia-reperfusion.METHODS: The ischemia model was established by blocking middle cerebral artery for 2 hours and reperfusing for 0. 5 -48 hours, and control sham operated(n=3) and normal(n=3) rats were processed. The immunocytochemical and lectin GSI-B4 by single and double staining were used to investigate temporal profile and morphological characteristics of microglia/macrophages after induction of focal cerebral ischemia in adult rats.RESULTS: There were many GSI-B4-positive microglias/macrophages with different morphologies as early as within 0. 5 hour. The number of positive cell declined from 3 to 6 hours and increased again from 12 to 48hours. The GSI-B4-positive macrophages were located in the superficial layer of preoptic area and cortical amygdaloid nucleus and meninges covering them. In control groups and all ischemia-reperfusion groups, PCNA positive immunohistochemistry was present in the ventricular and subventricular zones(VZ/SVZ) .Moreover, the strong PCNAimmunoreactivityandGSI-B4 strong positive amoebic-like macrophages were present in amygdaloid nucleus,cortical amygdaloid nucleus and meninges covering them. The double staining for PCNA and CD45RB existed the same cell from 24 to 48 hours.CONCLUSION: In ischemia-reperfusion, the microglia showed distinctly different morphologies. The extraneous macrophages may deride from not only the proliferation, immigration of macrophages in meninges, but also from the proliferation, immigration and differentiation of VZ/SVZ.
