Abstract
The callus formation ability of maize leaf segments decreased gradually as their positions from basal to middle and upper segments. It corresponded to the change of endogenous IAA and ABA. The callus formations of basal segments had no associated with either 2, 4-D or combinations of 2, 4-D and BA in media, and coincided with the Logistic curve. The BA alone had no effect on callus formation. The combination of 2 mg·L-1 2, 4-D and 0.5 mg·L-1 BA was recommendable for inducing the embryogenic callus. Endogenous levels of IAA in callus cultures reduced as the culture passed and the 2, 4-D concentration increased. Endogenous levels of ABA were enhanced by 2, 4-D, especially in 2 mg·L-1. As the endogenous level of ABA in the cultures reached to maximum value, the embryogenic calli formed. Therefore the occurrence of embryogenic callus might be relate to endogenous levels of ABA, which was affected by 2, 4-D.关键词
