Abstract
BACKGROUND: The failure in the establishment of focal cerebral ischemia model is usually due to nylon monofilament because of its lacking of a certain rigidity, which led to inadequate depth. Therefore, to find a thread,which has an internal homogeneity and certain rigidity and elasticity becomes a key to increase the successful rate in the establishment of rat focal cerebral ischemia model caused by middle cerebral artery occlusion (MCAO) using thread ligation.OBJECTIVE: To find a way to increase the successful rate in the establishment of rat focal cerebral ischemia model caused by MCAO.DESIGN: A randomised controlled study.SETTINGS and MATERIALS: The study was carried out in the Central Animal Laboratory of Neurology Department, General Hospital of Jinan Command. Sixty male Wistar rats with a body weight of 160 - 270 g were obtained from Experimental Animal Centre of Shandong Medical University.Rats ate freely before study. All the rats were randomly grouped into rat moustache and nylon monofilament groups.INTERVENTIONS: To compare the effects of rat moustache managed by polyvinyl alcohol and nylon monofilament on model successful rate and infarction volume.MAIN OUTCOME MEASURES: Model successful rate and infarction volume.RESULTS: The model successful rates of rat moustache and nylon monofilament groups were 96% and 55% respectively. There was no significant difference in the infarction volume 12 hours after ischemia[ (209. 6 ±21.4)vs (227.6 ±40. 9)mm3]CONCLUSION: Rat moustache has an internal homogeneity with good rigidity and elasticity. Good expansibility of polyvinyl alcohol can recuperate diametrical non-homogeneity in rat moustache. Therefore, rat moustache managed by polyvinyl alcohol can significantly increase the successful rate in model establishment.关键词
