转化生长因子-β1对瘢痕疙瘩成纤维细胞Smad3,7mRNA表达的影响OACSTPCD
Expression of Smad3 mRNA and Smad7 mRNA of TGF β 1 in keloid-derived fibroblasts
目的探讨转化生长因子-β 1( TGF β 1)对瘢痕疙瘩成纤维细胞 (KFB)Smad3,7 mRNA表达的影响,以进一步明确 TGF β 1在瘢痕疙瘩的发生、发展过程中的重要作用.方法用 RT-PCR法分别检测 TGF β 1 不同含量( 0, 5, 50, 500 pmol/L;Smad3 mRNA 24 h, Smad7 mRNA 90 min)和 TGF β 1( 500 pmol/L)作用不同时间( Smad3 mRNA: 1, 2, 4, 24, 48 h; Smad7 mRNA: 0.5,1,1.5,2,4,24 h)对 Smad3,7mRNA表达的影响.结果不同含量 TGF β 1刺激 KFB后 ,随 TGF β 1含量的增加 ,Smad3 mRNA水平逐渐下降,并呈含量依赖性.与 Smad3相反, 5 pmol/L TGF β 1就能使 Smad7 mRNA水平明显升高 2.6倍( 46± 7, 对照组 13± 7, t=6.150, P=0.0005),并随 TGF β 1剂量加大而略有改变,说明 KFB的 Smad7 mRNA表达对 TGF β 1的刺激非常敏感.用 TGF β 1刺激细胞后, Smad3 mRNA水平随作用时间延长而逐渐下降,到作用 48 h后下降到最低( 53± 9,对照组 18± 8, t=6.011, P=0.0005),显示出时间依赖性;而 Smad7 mRNA在细胞受到 TGF β 1刺激后 120 min即达到对照组的 2.5倍( 73± 11,对照组 53± 9, t=5.215, P=0.003) ,24 h后回落到正常对照水平.结论 TGF β 1能使 KFB细胞 Smad3 mRNA发生配体依赖性的下调,而使 Smad7 mRNA表达迅速增加.
Aim To study the expression of Smad3 mRNA and Smad7 mRNA of transforming growth factor β 1 ( TGF β 1) on keloid-derived fibroblasts. Methods The expression of Smad3 mRNA(at 1,2,4,24,48 h)and Smad7 mRNA(at 0.5 1,1.5, 2,4,24 h) were detected by using RT-PCR after TGF β 1 in different concentration(0,5,50,500 pmol/L) were added to the monolayer culture system. Results Expression of Smad3 mRNA decreased with increase of TGF β 1 in a concentration dependent manner.Contrary to Smad3,5 pmol/L TGF β 1 significatnly enhanced the expression of Smad7 by 2.6 time( 46± 7,control group 13± 7,t=6.150, P=0.0005) ,its expression increased with increase of TGF β 1 dose indicating Smad7 mRNA was very sensitive to TGF β 1 stimulation. Smad3 mRNA level decreased after TGF β 1 stimulation and reached the minimum value 48 hours later ( 53± 9,control group 18± 8,t=6.011, P=0.0005) and showed time dependence; Smad7 mRNA level was increased compared with control group ( 73± 11,control group 19± 10,t=5.215, P=0.003) ,24 h later,returned to the level similar to that in control group.Conclusion TGF β 1 may down regulate the expression of Smad3 mRNA,but up regulate that of Smad7 mRNA in a ligand-dependent way.
