Abstract
Objective: To investigate the effects of NGF and BME on differentiation of mMSCs into neuron-like cells. Methods: MMSCs were isolated by density gradient centrifuge with Percoll-paque (1.073 g/ml)and purified by adherence differention, and stimulated to proliferate by PDGF-BB and EGF. MMSCs were treated by nerve growth facor (NGF) and 2-mercaptoethanol (BME). The morphology changes were observed during induction. Immunocytochemistry analyses were used to detect the expression of NSE, NF and GFAP.The cells were stained with Toluidine Blue to show the Nissl bodies. Resulsts: Immunocytochemistry showed that NSE was expressed by 70% of the cells, and 74% of cells expressed NF. The induction group had the Nissl bodies. Conclusions: NGF and BME can induce mMSCs to differentiate into neuron-like cells and can get high induction efficiency.关键词
