Abstract
Honeybee (Apis cerana cerana) semen was stored for 121 days at -196 ℃ in liquid nitrogen.Samples consistng of semen and tris buffer diluent containing 8% dimethyl sulphoxide,at 2 or 5 cm over the liquid nitrogen surface in a broadmouth thermos for 5-10 min to prefreeze and at 35-39 ℃ water bath to thaw,could produce over 60% post-thaw motility rate without equilibration(the radio between semen and tris buffer diluent was 1∶1.5).When the room temperature went beyond 20 ℃ (in April),samples must be equilibrated at 13 ℃ for 0.5 h or so,the post-thaw motility rate of the freezing sample would be over 30%.The post-thaw motility of samples collected and frozen in spring were not so good as that in winter,twenty four virgin queens were inseminated instrumentally with 4-5 μL semen stored for 1-121 days in liquid nitrogen,four of these queens laid but didn,t lay fertilized eggs first.Frozen semen could be stored for 46 h at 13 ℃ after thawing.Sulphanilamide should not be used in semen freezing.关键词
