星形胶质细胞诱导神经干细胞定向分化试验研究
Experimental study on committed differentiation of neural stem cells induced by astrocytes
摘要
Abstract
[Objective]To investigate the effect of the culture medium of astrocyte on differentiation of neural stem cells (NSCs) in vitro, and explore the proper condition of neural stem cells' differentiation.[Method]The medium from cultured astrocyte of neonatal and adult SD rats were collected respectively and mixed with DMEM/F12medium according to the proportion of 1:3. Fetal brain derived NSCs were induced to differentiate in the mixed mediums. The induced cells were observed under the phase contrast microscope and were detected and counted by immunofluorescent technique.[Result]The mixed medium of neonatal astrocyte culture medium and DMEM/F12medium induced more NSCs to neurons. [Conclusion]The medium from cultured neonatal astrocyte promotes NSCs' neuronal differentiation.关键词
