摘要

Abstract

PAV Chinese isolate of BYDV isolated and purified from our national wheat.The coat protein(CP) gene of this isolate was identified by reverse transcription polymerase chain reaction(RT-PCR),clone and s equence.The cloned fragment contains a single open reading frame of 00 bp which encode 199 amino acid.PAV CP was cloned into expression ector pEmu-mcs-N to construct the plant expression vector pPPI10.In the term of the two kinds of cloned CP genes of PAV and GPV,we onstructed plant expression vector pPPI14 which contain CP genes of PAV and GPV,then transformed the two kinds of plasmids into wheat cultivar Beijing 837 by pollen tube pathway.Transgenic plants were confirmed by Kanamycin selection,dot blot,PCR and Southern blot assay,the transformation frequence is 2.5% and 0.36%,respectively.Most of transgenic plants showed reduction in virus accumulation observed upon virus infection.Compared with the control delay in symptom development BYDV resistance inoculation in the greenhouse and fields,so we can conclude that transgenic plants gained certain resistance.
