Changes of the contents of cerebral calcium in different areas after cerebral contusion in rats

Abstract
BACKGROUND: Intracellular calcium overload in neurocyte induces serials of pathophysiological reaction and therefore, causes injuries in neurocyte. The application of Ca2 + channel blocking agent in the treatment of craniocerebral trauma provides a theoretical gist.OBJECTIVE: To discuss the interference of Ca2 +-PO43 - in the measure of inductance coupling plasma atom emission spoctrum(ICP-AES) and to set proper analytical conditions for the assay of the changes of cerebral calcium content after cerebral contusion in rat.DESIGN: A randomized controlled study.SETTING and MATERIALS: The experiment was completed in Medical Chemical Laboratory of Analysis Department of Benghu Medical College. Eighteen male Wistar rats with a body weight from 200 g to 300 g obtained from animal department of our University were randomly allocated into Ca2 + control group and trauma group with 9 rats each.INTERVENTION: To study the impacts of different strength P(V) (0 -2 000 mg/L) at 5 observation heights on the intensity of Ca2+ spectrum; the impact of 2 000 mg/L of P(V) on Ca2 + when the current at positive pole changed; and the impact of P(V) on Ca2+ when air pressure changed. Cerebral intracellular free calcium strength after cerebral contusion was assayed by ICP-AES and the pathological changes were observed as well. ,MAIN OUTCOME MEASURES: The content changes of calcium at different cerebral areas after cerebral contusion.RESULTS: The analytical condition set in our study could significantly decrease the interference of PO43-, with the relative standard deviation(RSD% )of 1.16 for 20 times of assays and 97% of rate of recovery in marked samples. Cerebral intracellular free calcium significantly increased after cerebral contusion. Intracellular calcium contents at each cerebral area were significantly higher than that of control group( t = 3.45 - 46.39, P < 0.01 ).CONCLUSION: Cerebral contusion could lead to the openness of calcium channel on the membrane of cerebral neurocyte, which would cause great intra-flow of calcium ions to induce calcium overload. It is extremely important to optimize the experimental conditions of ICP-AES for the decrease of the interference and the increase of the analytical accuracy.关键词
