摘要

Abstract

Aim To study the influences of mild hyperthermia,subhypothermia on excitatory amino acids in focal ischemic brain tissue.Methods 91 rats were divided by random number table into ischemia 2 h reperfusion 0,1,2,3 h groups(O2R0,O2R1,O2R2,O2R3) under different cerebral temperature conditions(mild hyperthermia,normal temperature and subhypothermia) and false operation group,13 groups in total(n=7 in each group); Rat model of ischemial and reperfusion of middle cerebral artery blocked with wires was used to induce optional cerebral temperature and contents of Glu,Asp,Gly were detected by HPLC fluorescence method.Results Contents of Glu,Asp,Gly in mild hyperthermia group at different phases were obviously higher than those in nomal temperature or false operation group at corresponding phases.0 h:the contents in mild hyperthermia group were (4.3± 0.6),(9.4± 1.0),(3.4± 0.5) μ mol/g;in normal temperature group they were (3.0± 0.4),(7.0± 0.6),(3.0± 0.4) μ mol/g;in false operation group they were (2.4± 0.3),(4.8± 0.5),(1.7± 0.3) μ mol/g;F=8.17-174.17, P=0.005 2-0.000 1.The contents in subhypothermia group at different phases were obviously lower than those in normal temperature or mild hyperthermia groups (F=5.80-174.17, P=0.017 9-0.000 1); Within 3 h after reperfusion,contents of Glu,Asp and contents of Gly in subhypothermia and normal temperature groups decreased in different degrees,but content of Gly in mild hyperthermia group showed no obvious decreasing but was still higher than that in normal temperature group( F=8.57, P=0.004 3) .Conclusion Mild hyperthermia promotes increasing of EAA and plays an important role in continuous increasing of Glu exitotoxicity;Subhypothermia inhibits increasing of EAA and plays an important role in decreasing of Glu exitotoxicity.
