The Sequestered Actin Was Bound to Non-actin Protein (s) in the ATP-depleted Rabbit Proximal Tubules

Abstract
Objective: To demonstrate whether the actin breakdownfrom actin-cytoskeleton in microvilli was bound to any non-actin protein (s) in ATP-depleted rabbit proximal tubules(PT). Methods:By using DTSSP to crosslink actin and non-actin protein (s) in insoluble fraction of ATP-depleted PT, the crosslinked protein complexes were analysed by two-dimensional electrophoresis and western blotting. Results: Analysis of the crosslinked actin complexes revealed one complex that contained actin and non-actin protein. Villin was not detected. Conclusion : The results strongly suggested that some of the sequestered actin was directly bound to non-actin protein (s) other than was co-precipitated with villin or the nuclei as hydrophobic actin homopolymers .关键词
