青稞中β-1,3葡聚糖酶的纯化及部分性质研究
PURIFICATION AND ACTIVITY STAINING OF β-1,3-GLUCANASE FROM HIGHLAND BARLEY
摘要
Abstract
A β-1,3-glucanases [E. C. 3.2.1. 39] was isolated from seedlings of highland barley(Hardeumvulgare var. nudum Hook. F. ) by NaCl extract, ammonium sulfate precipitation, two-steps of ion-exchangechromatography and gel filtration on Sephadex G-100 column. The purified β-1,3-glucanase showed only a sin-gle band in native-PAGE both by activity staining and Coomassie Brilliant Blue R-250 staining. Under reducingand non-reducing conditions,it showed one major bands on SDS-PAGE corresponding to molecular mass of 32kD with two minor bands. After isoelectric focusing, the enzyme gave a band at pH 8.1 by activity stain, indi-cating that its pI was 8.1. Our results suggest that the purified β-1,3-glucanases was belong to an alkaline andmonomeric one.关键词
