Abstract

Objective To validate the human dendritic cell(DC) derived-exosomes(EXO) loaded with NY-ESO-1 effects of antigen in promoting the amplification of specific cytotoxic T lymphocytes (CTLs)in transgenic mice and enhancing the anti-tumor effect. Methods Peripheral blood mononuclear cells (PBMC) obtained from human leukocyte antigen (HLA)-A2-positive healthy adults were used to induce and isolate mature DC. A series of differential ultracentfifugation combined with ultrafiltration were used to isolate the EXO secreted by DC from the supematant. Then the EXO was loaded with testis cancer antigen NY-ESO-1 (EXO/Ag), and used in conjunction with polyinosinic acid-polycytidylic acid(Polyl-C) to immunize healthy HLA-A2 transgenic C57 mice. The spleen cells were cultured in vitro and the EXO/Ag was added to the system. Spleen cells were tested for tetranser staining and eytotoxicity against K562 cell and melanoma cell(control) killed. Results NY-ESO-1-specific CTLs were amplified and displayed cytotoxic effect on tumor cell line K562. In vitro study showed that NY-ESO-1-specific CTLs amplified substantially, and their killing effect on tumor cell line K562 was greatly enhanced. Conclusions EXO/Ag in conjunction with PoIyI-C has the prospect to be a new generation of anti-tumor vaccine.
