响应面法优化磁性固定化纤维素酶的制备
PREPARING MAGNETIC IMMOBILIZED CELLULASE BASED ON RESPONSE SURFACE METHODOLOGY

Abstract
The conditions such as immobilized time, pH value, buffer solution concentration and the amount of cellulase for preparing magnetic immobilized cellulase were optimized by means of the response surface methodology (RSM), the activity of magnetic immobilized cellulase was chosen as the response value. And the mathematical regression model between different variables and the activity of magnetic immobilized cellulase was obtained using regression equation. Determination coefficient (R2) was 0.902 4, lack of fit did not exist. The results demonstrated the optimum parameters of preparing magnetic immobilized cellulase were: immobilized time of 11.98 h, pH value of 5.2, buffer solution concentration of 0.066 mol·L-1 , the amount of cellulase used of 202.7 mL. Under the optimum parameters, the maximal activity of magnetic immobilized cellulase was 28. 457 44 IU·g-1.关键词
