Effect of indomethacin on the expression of Bfl-1, WISP-1 and PCNA in the colon cancer HCT116 cells

Abstract
[Objective] It has been known that non steroidal anti-inflammatorydrugs (NSAIDs) can inhibit the growth of tumors through both the COX-2 dependent and independent way. To study the effect of indomethacin on the expression of Bfl-1, WISP-1 and PCNA mRNA in the human colon cancer HCT116 cells and its COX-2 independent way to inhibit tumors. [Methods] Human colon cancer HCT116 cells were treated with different concentration of indomethacin for 48 h to obtain its IC50 (50% inhibitory concentration), then the cells was treated with IC50 for 48 h. The expression of Bfl-1, WISP-1 and PCNA mRNA was detected by real-time quantitative PCR. [Results] The IC50 of colon cancer HCT116 is 316μmol/L indomethacin. Indomethacin down-regulates mRNA expression of Bfl-1, WISP-1 and PCNA in vitro. [Conclusion] Inducing apoptosis and inhibiting proliferation contribute to the anti-tumor activity of indomethacin by the COX-2 independent way of Bfl-1, WISP-1 and PCNA.关键词
