发根农杆菌Ri质粒转化康乃馨初步研究
A PRELIMINARY STUDY ON CARNATION BY RI PLASMID OF Agrobacterium rhizogenes

Abstract
Using Ri plasmid of Agrobacterium rhizogenes (Pri15834), thecarnation transformation experiment with leafed stem segments of shoot from tissue culture as initial material was carried out. After transformation, explants were transferred to K0 medium with 500mg/L carbenicillin and cultured for 2~3 weeks, then non-geotropic hairy roots formed from infected wound sites at a frequency of 37.07%. Afterwards,the hairy roots were cut up and subcultured on K0,K1,K2 mediums with 250mg/L carbenicillin for 3 weeks. The results of subculture showed that the hairy roots on K0 medium grew indefinitely, those on K1 medium formed calli at a frequency of 51.42%, those on K2 medium formed green plantlets at a frequency of 25%, indicating expression of Ri plasmid integrated into the genome of carnation cell.关键词
