Abstract
AIM:To investigate the relationship between human prostate cancer cells with different metastatic potential and expression of COX-2. METHODS: Expression of cyclooxygenase-2 and it's regulation by tumor necrosis factor-α in PC3 and PC3M were detected by reverse transcription-PCR and immunohistochemistry. RESULTS:We observed that the COX-2 mRNA level and COX-2 protein were undetectable in PC3 cells but it was positive at 4 h after stimulation with TNF-α .In PC3M, there were a gradual increase in COX-2 expression up to 4 h after stimulation with TNF-α compared with the beginning. CONCLUSION: The result of the current study demonstrate that the human prostate carcinoma cells generated COX-2 and regulated by TNF-α ,and that COX-2 might play an important role in the proliferation of prostate carcinoma cells.These findings suggest that inhibition of COX-2 development may lead to inhibition of the proliferation and metastasis of prostate carcinoma but also to the inhibition of prostate carcinogenesis.关键词
