甜荞愈伤组织分化研究
Study on Callus Differentiation of Common Buckwheat*

Abstract
Calli from cotyledon and hypocotyl explants of four cultivars of common buckwheat were induced on Murashige and Skoog (MS) basal medium and the effects of plant growth regulators and sucrose concentrations on callus differentiation were investigated.The results showed that the combination of 17.8 μmol/L~26.6 μmol/L(4.0 mg/L~6.0 mg/L) of 6-benzyladenine (BA) with 0.9 μmol/L(0.2 mg/L) of 2,4-dichlorophenoxyacetic acid (2,4-D) was effective in inducing differentiation and generation of adventitious buds.Comparative studies using different sucrose concentrations (1.5 %~6.0 %) demonstrated that higher sucrose concentrations (4.5 %~6.0 %) promoted root differentiation and enhanced callus growth.Addition of activated charcoal into the culture media inhibited root differentiation.关键词
