Abstract
BACKGROUND:Although researchers home and abroad have paid attention to studies on brain damage possibly caused by febrile convulsion, studies of effects on behavior motion, learning and memory are relatively seldom.OBJECTIVE:To study the effects of repeated febrile convulsion on behavior motion, spatial learning and memory in rats.DESIGN:Randomized controlled experiment.SETTING and PATICIPANTS:All experiments were performed in the Laboratory Animal Center of Xi' an Jiaotong University.Sixty 21-day-old male Sprague-Dawley rats were randomized into febrile convulsion group (FC, n = 20), hypothermic control group (HC, n = 20), normal control group (NC, n = 20).INTERVENTIONS:FC model was induced by hyperthermal bath with 45 ℃water.HC model which rats were bathed with the same water in fixed 2 minutes was also developed in this study. The abilities of motor and behavior were tested by inclined plane test (IPT), overhanging test(OHT) and open field test (OFT) .Morris water maze(MWM) was applied to show the variations in abilities of spatial learning and memory in each group.MAIN OUTCOME MEASURES:The indexes included turning time, overhanging time, activity, escape latency and search strategy.RESULTS:Repeated febrile convulsion could lead to decreases of abilities in IPT[(9.15±2.63) s for FC, (5.32±2.11) s for HC and(5.29±2.04) sfor NC.FC vsHCorNC, F=6.17, P <0.01], OHT[(33.4±18.1) sforFC,(50.1±20.3) sforHC and(59.0±20.7) s forNC.FC vs HC orNC, F=8.23, P < 0.01 ] and OFT[ (5.07 ± 2.04) scores for FC, (10.37 ± 2.98)scores for HC and(13.22 ± 2.26) scores for NC, FC vs HC or NC, F = 7.42,P < 0.01 ] .In MWM test, the escape latency prolonged, the percentage of swim time in platform quadrant reduced and the times of swimming across platform area decreased in FC group.CONCLUSION:Febrile convulsion can impair behavior motion, spatial learning and memory in rats.关键词
