Abstract
AIM:To observe the therapeutic effectiveness of acetyl-Asp-Glu-Val-Asp-ald-ehyde (AC-DEVD-CHO), a specific Caspase-3 inhibitor, in the treatment of hypoxia-ischemia brain-damaged(HIBD) neonatal rat models.METHODS:Different dose of AC-DEVD-CHO was administrated through cerebral ventricular injection in the neonatal rats after the establishment of HIBD model.The activities of Caspase-3 in the brains were measured after 24 hours by four-peptide-fluorescence-substance detection to determine the optimal dose.The therapeutic effectiveness of AC-DEVD-CHO was observed by means of histopathology and in situ apoptosis test, etc.RESULTS:The activity of Caspase-3 descended with the increasing dose of AC-DEVD-CHO.The difference was not significant when the dose reached 300 to 400 ng.The percentage of apoptotic neurocytes(30.2 ± 11.2%) in the cerebral cortex and hippocampus of HIBD neonatal rats in AC-DEVD-CHO 300 ng group was significantly lower than that(44.6 ±12.4%) of normal saline group( P < 0.01).CONCLUSION:Caspase-3 inhibitor has cerebral protective effect in HIBD neonatal rats.关键词
