Abstract

The farming of the Chinese mitten crab, Eriocheir sinensis, has been badly struke by the epidemic of tremor disease (TD) during the last few years. The proPO system is the most important humoral defense system in crustaceans. When the pathogens invade the crab's body, it performs basic immune response against infection. The change of the proPO system is an essential marker for crab's health status. The prophenoloxidase and peroxinectin (pero) are two important factors in the proPO system. The aim of this study was to develop a method to determine the crabs' health by detecting the proPO system at both protein level and molecule level. We designed degenerate primers based on the highly conserved nucleotides of proPO and pero of other decapods. 203 bp cDNA sequence of proPO and 296 bp of pero isolated from haemocytes of E. sinensis were cloned by RT-PCR with the degenerate primers. Sequence analysis with the BLAST algorithm showed that the two nucleic acid sequences of E. sinensis proPO and pero exhibited the high similarity to those of other decapods. In addition, we also compared the change of the proPO system in normal crabs with those infected with TD. The result showed that the PO activity increased in protein level when infected with TD. In molecule level, the mRNA expressions of the two factors were on a very low level in the normal group, whereas the infected group, their expressions increased dramatically. These evidences suggests the proPO system executes an important function in host immune response and might bring a new way to deal with TD infection.
