复方黄连处理后HNE1鼻咽癌细胞移植瘤的基因表达
Gene expression in tumor tissue transplanted with HNE1 nasopharyngeal carcinoma cell after treatment with Coptis chinensis

Abstract
BACKGROUND: The compound of Coptis chinesis can inhibit the growth of tumor transplanted with nasopharyngeal carcinoma(NPC) cell, but its mechanism remains not fully clear.OBJECTIVE: To investigate the mechanism of the inhibitory effect of compound Coptis chinensis on the growth of transplanted tumor with NPC cell.DESIGN: A randomized controlled trial study was conducted.SETTING, MATERIALS and METHODS: The experiment was completed in Animal Experiment Center of Xiangya Medical College, Central South University. Ten Balb/c nude mice of 3 - 4 weeks old were selected. After feeding for one week, 0. 5 x 107 /0. 5 mL HNE1 cells were injected into the front limb of each nude mouse. Once the growth of tumor was visible, the nude mice were divided into experimental group and control group with 5mice in each group according to the size of the transplanted tumor. The experimental group was treated with Coptis chinensis compound while the control group was only perfused with sterile water. RNA were isolated from the tumor tissues, hybridization with a cDNA microarray was performed with a probe consisting of a mixtureof fluorescent-labeled cDNAs reverse-transcribed from RNA, and gene expression was analyzed by scanning the fluorescent intensity and further confirmed by RT-PCR.MAIN OUTCOME MEASURES: Gene expression in tumor transplanted with HNE1 cell of NPC after treatment with Coptis chinensis compound.RESULTS: After treatment with Coptis chinensis for 30 days, the transplanted tumors were remarkably reduced in size. Meanwhile, expression of 335 genes was altered, of which 242 were down-regulated and 93 were up-regulated. Altered expression of 6 of these genes was confirmed by RT-PCR. Among them, mRNA expressions of the MAD3, H731 and EGR-αgenes were increased, but those of TRAF5, p68 kinase and CHK1 were decreased.CONCLUSION: The compound of Coptis chinensis can inhibit the growth of tumor implanted with HNE1 cell of NPC in nude mice, which may be associated with the change in gene expression.关键词
