氯沙坦对糖尿病大鼠肾脏蛋白激酶C活性作用的研究
Effect of losartan on renal protein kinase C in diabetic rats
摘要
Abstract
[Objective] To investigate the preventing effects and possible mechanism of angiotensin AT-1 receptor blocker (ARB), losartan on renal protein kinase C(PKC) in diabetic rats. [Methods] Experiment diabetic ratswere induced by peritoneal injection of streptozotocin. The rats were randomly divided into following groups: normalcontrol (group C),streptozotocin diabetic rats (group D) and diabetic rats treated with losartan (group DL). After twen-ty-four weeks, glomeruli were isolated. ①Subcellular fraction prepared and partially purified. Subcellular distribu-tion of PKC was detected by using [γ-32P] ATP as the phosphorus donor; ②The concentration of serum and urinecreatinine or the creatinine clearance rate was also determined. [Results] After 24 weeks, diabetic rats had obviousrenal hypertrophy. The activity of PKC in cytosolic fraction was found significantly decreased in glomeruli from dia-betic rats as compared to the control. Total cellular PKC activity was not significantly changed. The PKC in mem-brane fraction and the percentage of PKC activity associated with the membrane fraction of glomeruli from diabeticrats were found significantly increased, meanwhile, losartan can prevent the increases of PKC activity in glomeruli of diabetic rats. Creatinine clearance rate or 24-h urine protein of group DL was lower than that of group D. [ Con-clusion] Losartan ossesses a renal protective effect on diabetic rats, whose mechanism may be associated partly with a down-regulation of PKC activity.关键词
