Abstract
BACKGROUND: It is observed clinically that yiqi huoxue fang(Recipe)prevents against radioactive bone injury. But the mechanism of it is unclear yet at present.OBJECTIVE: To probe into the preventive mechanism of Recipe against radioactive bone injury.DESIGN: A randomized controlled experimental study.SETTING and PARTICIPANTS: The experiment was accomplished in Center for Laboratory Animals, the 251 Hospital of PLA and Center for Laboratory Animals, Third Military Medical Hospital. Forty-eight Japanese flap-eared white rabbits were employed in either gender, weighing from 2.0to 3.0 kg, aged from 8 to 12 months.INTERVENTION: Forty-eight rabbits were randomized into experimental group and control group, 24 rabbits in each. All of those were radiated once only with 60Coγ-rays at 30 Gy and the model was prepared on hind legs. All the herbs were decocted and sediment, each milliliter contained raw herbs 2 g. In the experimental group, gastric infusion was done daily with Recipe at 2 mL,composed of Huangqi(Radix Astragali seu Hedysari) 30 g, Taizishen(Radix Pseudostellariae) 30 g, Baizhu(Rhizoma Atractylodis Macrocephalae) 15 g,Fuling(Poria) 15 g, Danshen(Radix Salviae Miltiorrhizae)30 g, Jixueteng (Caulis Spatholobi) 30 g, Dilong(Lumbricus) 15 g, Chishao(Radix Paeonine Rubra ) 30 g and Gancao (Radix Glycyrrhizae) 10 g. Since radiation, the Recipe was given for 6 months. In the control, the rabbits were infused with normal saline at the same dosage.MAIN OUTCOME MEASURES: To observe vascular cast and blood flow on the radiated region in the 1st, 3rd, 6th, 12th, 24th and 52nd weeks after radiation successively.RESULTS: The degenerative necrosis of osteocytes was developing, which was severer in the 6th week, 27% in the control and 8% in the experimental group, indicating remarkable difference. Concerning to the vascular cast, the vascular nets were increased between the 1st and 3rd weeks, which were of vesicular dilatation and fusion into patches. Afterwards, they were decreased gradually. In the 24th week, the numbers of vessels in the experimental group were remarkably more than those in the control In the 6th, 12th, 24th and 52nd weeks, the local blood flow volumes in the controls were (3.76 ± 0.28),and were (4.46 ± 0. 35), (3.73 ± 0. 27), (3.06 ± 0. 21), (3.10 ± 0.27)L/there was very significant difference ( t = 8. 6549, P ＜ 0.01 ).CONCLUSION: The Recipe can weaken the direct injury of rays on osteocytes and other tissues, reduce bone vascular injury and improve bone microcirculation and blood flow.关键词
