Abstract
BACKGROUND:Researches find that Alzheimer disease(AD) may relate with the pathological gene in chromosome ;However, therelationship between Apolipoprotein E(ApoE) gene that located on No. 19 chromosome and the onset of AD and vascular dementia (VD) is still not clear.OBJECTIVE: To evaluate the relationship between ApoEe4 gene and AD and VD for the discussion of its application merits in the evaluation of prognosis. DESIGN: A non-rnndomised and controlled study based on diagnosis.SETTING and PARTICIPANTS: A total of 1 560 ageing patients from the outpatient Department of Dysmnesia of BROCA Hospital (Paris, France)were selected from January 1996 to December 2000. All cases were Caucasians, which were divided into three groups: non-dementia group( n = 845,patients with normal cognitive function or mild cognitive dysfunction ), Adgroup( n =618) and VD group( n =97).MAIN OUTCOME MEASURES: The detective rate of ApoE alleles of each group, analysis of the independent risk factors of AD and VD.RESULTS: A total of 309 cases in AD group (50%) carried E4 alleles,which was significantly different from that of 240 cases in non-dementia group( P < 0.01).Multiple factors were analysed byregression analysis with the results of that ApoEe4, age and female are the independent risk factors of AD; However, ApoEe4 is not the independent risk factor of VD( P =0. 69).CONCLUSION:ApoEe4 significantly correlates with AD, which is an important risk factor of AD genesis;However,the relationship between ApoEe4 with VD is still unclear.关键词
