Separation, Purification and Component Research of an Extracellular Polysaccharide Produced by Trichoderma Pseudokoningii SDTP1

Abstract

A water-soluble extracellular polysaccharide (TPP-0) was obtained from Trichoderma pseudokoningii SDTP1, by deproteination using enzymolysis-Sevag method, and fractionation through Sephadex G-100 chromatography. Monosaccharide composition and the preliminary structure, molecular weight of TPP-0 were studied by ultraviolet (UV), infrared (IR), gas chromatography (GC), and high performance liquid chromatography (HPLC). TPP-0 had characteristic UV absorption peaks, being identical with normal polysaccharide. High performance gel permeation chromatography (HPGPC) was used to determine the molecular weight of TPP-0, and the result showed that the average peak molecular weight of TPP-0 amounted to 1.8×104. Three monosaccharides, i.e. rhamnose, glucose, galactose were detected by gas chromatography from the TPP-0 sample. And TPP-0 also contained glucuronic acid. The molar ratio of three monosaccharides was measured.
