Abstract

AIM: To investigate the behaviour and histopathological effects of hypothermia after 30-minute of cerebral ischemia in rat and compare with the immediate hypothermia after cerebral ischemia.METHODS: Cerebral ischemia rat model was introduced and ischemic time was 10 minutes. Animals were randomly allocated into 4 groups:sham-operated group, normothermia group, delayed hypothermia group and immediate hypothermia group with 7 rat each. Open field behavior examine was carried out on the 5th day and hippocampus CA1 area histopathological examine was carried out on the 7th day after cerebral ischemia.RESULTS: The numbers of grid got over by normothermia group at 10minutes were 651 + 108, which were obviously more than that of in sham-operated group(278 ± 67), delayed hypothermia group(478 + 89) and immediate hypothermia group(368±46) (t=7.76, 2.21, 6.37, P <0.01-0.05) . And the numbers in delayed hypothermia group were distinctly more than that of sham-operated group and immediate hypothermia group( t =4.75, 2.90, P < 0.01 - 0.05) . The neuron numbers in hippocampus CA1 area(expressed by percentage of normal hippocampus live neurons, ie. live neurons of each group compared with sham-operated group):the numbers of neuron in delayed hypothermia group[ (42 + 7)%] were more than that of normothermia group[ (5 + 1 ) % ] ( t = 13.00, P < 0.01 ), but less than that of immediate hypothermia group[ (66 +10)% ] (t=5.20, P <0.01). Counting of neuron in the central CA1 area of hippocampus: more in delayed hypothermia group[ (60 +9)%] than that of normothermia group [( 10 ± 4)%] (t = 13.20, P < 0. 01 ), but less than that of immediate hypothermia group [(77 + 16) %] (t = 2.45, P < 0. 05 ). Counting of neuron in the lateral CA1 area of hippocampus: numbers in delayed hypothermia group [(71 ±: 13)% ] had no difference compared with that of immediate hypothermia group[ (80 ± 14)% ] ( t = 1.25, P > 0.05), but more than that of normothermia group[ (23 ±5)% ] ( t =9.14, 10. 14, P < 0.01).CONCLUSION: Delayed hypothermia can alleviate neural dysfunction and neuron necrosis in hippocampus after cerebral ischemia but it function is not as good as immediate hypothermia.
