Abstract

AIM: To study the effects of hypoxic ischemic brain damage(HIBD) on synaptic quality in the frontal lobe cortex of newborn rats.METHODS:Eighty seven-day-old Wistar rats were used in the experiment,the rat neonates at a litter were divided into the experimental group (E) and the sham operation group (C) by the method of lots. The models of HIBD were made of group E, in which the rat pups' right common carotid was separated through the median incision in the neck and then ligated with No. 1thread with ether anaesthasia. After 2 h preservation at a constant temperature, the rates were placed in a container with a mixture of nitrogen and 8%oxygen for 2 h. In the sham operation group only the right lateral common carotid was separated without ligation or treatment of hypoxia. The synaptic quality, numbers of the neurons and density of the nerve fibers of the frontal lobe cortex were studied by the methods of morphology and morphometry and followed up in 8 separate intervals.RESULTS: The results showed that in the experimental group, the spaces around neurons, neuroglia and blood capillaries became enlarged, many vacuoles appeared in the neuron, mitochondria were swollen and their ridges were indistinct, the rough endoplasmic reticulum swelled and degranulated with 6 h -7 d. Shriveled, irregular and even dissolved nuclei were also seen especially at 24 h and 72 h. Density of neurons and nerve fibers decreased significantly( P < 0.05, P < 0. 01) . The morphometric parameters of the synapse, surface density(Sv) and numerical density of surface area(Na) decreased with thinning of the dense layer of postsynaptic membranes( P <0.05, P < 0.01).CONCLUSION:The results indicate that the change following HIBD in the synaptic quality and quantity of neurons of the frontal lobe is one of the important factors which affect the development of intelligence.
