Effects of electroacupuncture on learning and memory abilities and the apoptosis of cerebral cells in rats with vascular dementia

Abstract
BACKGROUND: Although a specific and satisfactory therapeutic method for vascular dementia(VD) has not been developed, it is important to prevent VD by effectively avoiding risk factors, preventing cerebrovascular accident (CVA)or preventing the recurrence, development and complications in patients with CVA. Exploring the effective mechanism for VD treatment with electroacupuncture method is one of the hot issues in the study of VD therapy.OBJECTIVE: To observe the effects of electroacupuncture on learning and memory abilities and the apoptosis of cerebral cells in rats with VD.DESIGN: A randomized and controlled experimental study.SETTING: Acupuncture and Massage Vocational College and College of Basic Medical Sciences, Guangzhou University of Traditional Chinese Medicine.MATERIALS: Sixty female and male(half-half) healthy adult SD rats,weighing 200 - 250 g, were provided by the Research Center of Laboratory Animals, Guangzhou University of Traditional Chinese Medicine.METHODS: VD model of four-vessel occlusion was made in healthy SD rats from the Experimental Animal Center, Guangzhou University of Traditional Chinese Medicine. Morris water maze tests were used for place navigation test and spatial probe test. The apoptosis of nerve cells in the cerebral cortex and hippocampus were also tested with TUNEL method.RESULTS: The escape time ( latency ) of rats in model group ( 58.08± 38.77) s was significantly longer than that in sham-operation group(26.50 ±29.4) s in place navigation test; and they did not swim more times in platform quadrant than in the other three quadrants in spatial probe test. The number of apoptotic nerve cells in the cerebral cortex and hippocampus were 41.60 ± 3.71 and 35.83 ± 7. 11 respectively. The latency of the rats in electroacupuncture group was(29. 89 ± 32.62) s in place navigation test. In spatial probe test, the electroacupuncture group rats swam more times in platform quadrant than in the other three quadrants. There was no significant difference between model group and sham-operation group. The number of apoptotic cells in the cerebral cortex and hippocampus were 12. 84 ±5.44 and 11.30 ±4. 16 in sham-operation group, but the number was obviously smaller than that in the model group.CONCLUSION: Electroacupuncture can improve learning and memory ability in VD rats, and it can also markedly inhibit the apoptosis of nerve cells of the rats.关键词
