Abstract

OBJECTIVE: To study the enzyme kinetics ofβ-lactamase extracted from Escherichia coli and Klebsiella pneumoniae. METHODS: 8 strains of β-lactamases-producing E. coli and K. pneumoniae were collected from clinical specimens in our hospital in 2008. Antimicrobial susceptibility was tested by Kirby-Bauer method. Michaelis constant (Km) and maximal velocity (Vmax)were obtained by L-B Plot. RESULTS: 8 strains were all resistant to the β-lactam antibiotics and highly susceptible to carbapenem.The penlcillins, the first generation cephalosporins were hydrolyzed by all β-lactamase extracted from E. coli and K. pneumoniae.But the second and third generation cephalosporins were hydrolyzed by all β-lactamase extracted from E. coli and K. pneumonia in different degree. CONCLUSION: Carbapenem is the first choice for the treatment of infections induced by β-lactamases-producing strains. There are significant differences among hydrolysate profiles of β-lactamases produced by different β-lactamases-producing strains.
