Determination of the Concentration of Docetaxel in Human Plasma by HPLC

Abstract
OBJECTIVE: To establish a method for the concentration determination of docetaxel in human plasma. METHODS: Using the external standard method, plasma samples were extracted with acetonitrile, shaken, centrifugated and filtrated.Kromasil C18(250 mm×4.6 mm, 5 μm)column was adopted with mobile phase consisted of acetonitrile at flow rate of 1.0 L·min-1.The detection wavelength was set at 230 nm. Column temperature was room temperature, sensitivity was 2.0AUFS and injection volume was 10 μL. RESULTS: The linear range of docetaxel was 0.167~3.333 μg·mL-1(r=0.995 2). The detection limit for docetaxel was 0.167 μg·mL-1. The extraction recovery was ranged from 84.7% to 87.5%. The RSD of inter-day and intra-day were both lower than 10%. CONCLUSION: The method is simple, accurate and rapid for the plasma concentration monitoring of docetaxel.关键词
