Abstract

OBJECTIVE: To optimize the preparation technology of Amomum villous oil-hydroxypropyl-β-cyclodextrin (HP-β-CD) inclusion complex, and to identify the inclusion complex. METHODS: With the utilization ratio of volatile oil, ratio of oil content to the highest oil content and yield of inclusion complex as index, the stirring method, ultrasonic method and trituration method were optimized. The preparation technology of inclusion complex was optimized by orthogonal experiment taking the comprehensive evaluation score as index with ratio of volatile oil to HP-β-CD, concentration of HP-β-CD, inclusion time and inclusion temperature as factors. Microscopic identification method, DSC method, UV method and GC-MS method were used for the identification of inclusion complex. RESULTS: Ultrasonic method was the best preparation method, and the optimal preparation condition as following: the ratio of volatile oil to HP-β-CD was 1: 10 ( V: m), the concentration of HP-β-CD was 40%, the time and temperature for inclusion was 80 min and 55℃.No obvious change was found before and after inclusion. CONCLUSION: The optimized preparation technology of A. villous oil-hydroxypropyl-β-cyclodextrin is feasible and serves as reference for the production and research.
