Abstract

Using linear HPMC (hydroxypropylmethyl cellulose) and NIPAAm (N-isopropylacrylamide) as raw materials, the temperature-sensitive hydrogels HPMC/PNIPAAm were prepared via the semi-IPN technique.The prepared hydrogels were characterized by Fourier transform infrared (FT-IR) spectroscopy, and their interior morphologies were observed by fluorescent microscope. The thermo-sensitivity, equilibrium swelling ratio and swelling dynamics of the prepared hydrogels were investigated, and the 5-fluorouracil was selected to be used as the model drug for examining the controlled drug release property of the prepared hydrogels. The results show that the swelling ratios of the hydrogels decrease with increasing temperature; and it was found that, the drug release rate of the hydrogel can be controlled by adjusting its HPMC content and the temperature of releasing. In pH 7.4 buffer solution (37℃), the prepared hydrogel PNI-H01, which was prepared with the weight ratio of HPMC to NIPAAm being 1/100, has the maximal swelling ratio and the fastest drug release rate.The obtained results suggest that the HPMC/PNIPAAm hydrogel would be a potential thermo-sensifive gel for oral drug delivery.
