Abstract

Poor cold - resistance of centipedegrass is one of the major factors limiting its wide application. The objective of this study is to obtain cold - resistant somatic mutants from the seed of ‘ Cuilù No. 1 ' centipedegrass by induction and screening at low temperature. The callus induced by seeds of centipedegrass was cultivated at the low temperature of 0℃ for twenty - four days during the process of subculture and three pieces of survival callus tissues were obtained. somatic mutant is obtained after survival callus subculture proliferation, differentiation, rhizogenesis as well as transplantation. Somatic mutant and the control. LT50of were -6.946℃ and - 5.851℃, respectively, which was significant difference. The results of SRAP markers demonstrated that there is characteristic bands at 200 bp of somatic mutant and indicated that the somatic mutants of centipedegrass were genetically stable comparing with that of the control plant on molecular level. This experimentation may provide effective means for screening of cold - resistance somatic mutant of plants.
