四水系中华绒螯蟹天然群体遗传特征的微卫星标记分析
The population genetic structure analysis on four stocks of Eriocheir sinensis from different water systems using microsatellite markers

Abstract
Microsatellite molecular markers were used to analyze the population genetics characterizations of 3 stocks of Eriocheir sinensis collected from different water systems (Yangtze River, Liao River, Ou River) and 1 stock of F2 generation of Rhine River population.The results showed that the genetic diversity indices of the 4 stocks are high with average expected heterozygosities (He) as 0.741 1, and that the average observed heterozygosities (Ho) of the four stocks were higher than the average expected heterozygosities (He) in the all 6 microsatellite loci respectively.The average observed heterozygosities (Ho) of the four stocks were 0.916 7, 0.958 4, 0.930 6 and 0.799 5 respectively, and the average expected heterozygosities (He) were 0.7496, 0.7416, 0.758 5 and 0.714 9 respectively.The Ou River stock had the highest He, while the stock of F2 generation of Rhine River population had the lowest value, but there was no significant difference among the four stocks (P>0.05).The fixation indices (Fis) were used to evaluate the inbreeding or outcrossing of the 4 stocks, and the negative values were observed in the almost all 6 microsatellite loci in the 4 stocks.The genetic distance (Ds) between the every two stocks ranged from 0.068 8 to 0.174 3.The Ds between the stocks of Yangtze River and Liao River was closer (0.068 8) than any others and it can be inferred that the genetic exchange between them was more deeply.The values of gene flows (Nm) among 4 populations were all above 1, especially the gene flow between the stocks of Yangtze River and Liao River or F2 generation of Rhine River stock was 20.362 9 and 14.385 1 respectively, which suggests that the population of Yangtze River has had a frequent gene flow with the two others before.Cluster analysis showed that the Yangtze River, Liao River and F2 generation of Rhine River stocks were allocated in the same cluster, and Ou River stock was allocated in another cluster.Analysis of molecular variance (AMOVA) showed that genetic differentiation index of all the four populations was 0.040 3, of which the majority (95.96%)existed within each population, and the rest existed among 4 stocks (4.04%).The results suggested that genetic admixture of E.sinensis in the current waters are serious.关键词
