Abstract

The optimal cultivation time of seed for rapamycin fermentation was investigated. It shows that the highest yield of fermentation with seed liquor can be achieved at 60 h. The effects of different medium components on rapamycin production were examined systematically, and one optimal fermentation medium was formulated. Under these optimal fermentation conditions, the yield of rapamycin is improved from 95 mg/L to 170 mg/L, achieving a big improvement by 79% with the above optimization. The systematic research of rational screening techniques finds an applicable way to generate high-yield strains from original microorganisms. The highest yield reaches 420 mg/L, about 3 times higher than that of original strain.
