摘要
Abstract
Objective To observe the protective effects of panax notoginseng saponins on immunological liver injury in mice. Methods The model of mouse immunological liver injury was induced by injecting bacillus calmette guein (BCG) and lipopolysaccharide (LPS) into tail vein. Visceral parameters of liver and spleen were determined; levels of ALT and AST in serum, the activity of SOD, GSH-Px and the of MDA concentration were assayed by spectrophotometry; the levels of TNF-α and IL-lβ were detected by ELISA; the coefficient of liver were assayed and dimensions of hepatic damage in the histopathology were observed; TGF-β1 and NF-kB p65 in hepatic tissues were determined by the method of immunohistochemistry; Liver samples were collected after the experiment and stained by immunohisfochemistry of TNF-amRNA. Results PNS was found to significantly decrease coefficients of liver spleen(P<0.05, P<0.01 ), also decrease the serum transaminase activity, MDA and prevent the reduction of SOD and GSH-Px activities in liver homogenate and also effectively prevent the levels of TNF-α and IL-1β; histopathological observations also showed that PNS could ameliorate the liver injury in different degrees and were correlated with the biochemical parameters and that PNS could effectively prevent the expressions of TNF-β1 and NF-κB p65; moreover PNS significantly reduced TNF-amRNA in liver. Conclusion PNS has protective effects on immunological liver injury in mice.关键词
