摘要

Abstract

The GPV/GSgulang/09 strain of goatpox virus(GPV) was isolated from lung tissues of Small-tailed Han sheep suffered from pox disease by inoculating into Vero cells. PCR-RFLP and sequence analysis of p32 gene were conducted for classifying this virus. Two bands,which are specific to goatpox virus,were observed after the p32 gene was amplified from genomic DNA of GPV/GSgulang/09, and then digested with the endonuclease of Hinf Ⅰ . Three nucleotide bases for encoding residue of aspartic acid which are located at 166 to 168 positions of p32 gene of sheeppox virus (SPV) were absent in that of GPV/GSgulang/09. Comparing the sequence of the p32 gene amplified from GPV/GSgulang/09 strain with those available in GenBank,the results showed that GPV/GSgulang/09 shared the homology of 98. 1% to 98.3% with SPV and 99.2% to 99. 9% with GPV, respectively. In the phylogenetic analysis, GPV/GSgulang/09 was more closely related to GPV strains and clustered into GPV sub-branch of phylogenetic tree constructed based on p32 gene. These results showed that the outbreak of sheep pox associated with GPV occurred for the first time in China.
