Abstract
Objective: To observe the effects of passive smoking on ovarian ERα and PR protein and mRNA of Wistar rats,which can provide theorial evidence promoting of reproductive age women to avoid passive smoking. Methods: A model of rat smoking was built up. Healthy female Wistar rats with 200-250 g weight were divided into a control group and a smoking group at random The experimental rats smoked for 3 months and the control rats fed normally. After 3 months, all rats were sacrificed The expressions of ERα and PR protein were detected by immunohistochemistry and Western blot. The expression of ERα and PR mRNA was detected by RT-PCR Results: Immunohistochemical results showed that ERα positive material was mainly distributed in the cell nucleus of the ovarian granulosa cells. ERα expression was significantly lower in the ovary of the smoking rats than that in the control group. PR positive material mainly was distributed in the cytoplasm and cell nucleus of the rat ovarian granulosa cells. PR expression was significantly lower in the ovary of the smoking rats for three months than that in the control group. Westem blot results showed that expression of ERα and PR protein in ovary of passive smoking rats was significantly lower than that in the control group. RT-PCR results showed that expression of ERα and PR mRNA in the ovary of passive smoking rats was significantly lower than that in the control group. Conclusiou : A rat model of passive smoking was successfully established Passive smoking can decrease the levels of ERα and PR protein, and down-regulate the expression of ERα and PR mRNA in the rat ovary. The results show that passive smoking can damage ovarian function and cause disorders of reproductive endocrine.关键词
