Abstract
BACKGROUND: For studies of some drugs, mouse serves as an ideal animal for model establishment. However, the failure rate is high due to poor toleration, small size of the kidney and renal pedicle.OBJECTIVE: To construct the model of renal ischemia/reperfusion injury (IRI) in BALB/c mice and evaluate influences of different ischemic time on IRI.METHODS: Renal arteries of mice were bilaterally clamped with micro-artery clamps to establish model of renal IRI in male BALB/c mice. According to different ischemic time, the mice were divided into four groups: 0 (control), 30, 35, and 45 minutes. The animals were sacrificed at 24 hours post-operatively. Renal function and pathology of the kidneys were examined. The situation of illness and survival in 45-minute group were observed.RESULTS AND CONCLUSION: The successful rate of model was 95.9%. Levels of serum creatinine and blood urea nitrogen as well as histological scores were remarkably increased in 30-, 35-, and 45-minute groups compared with control group 24 hours after operation (P < 0.05). However, the survival rate was significantly lower in 45-minute group (P < 0.05). Results show that stable model of renal IRI can be gained in BALB/c mice by applying micro-artery clamp to incarcerate bilateral renal arteries. The optimal renal ischemia time is 35-45 minutes in male mice.关键词
